The game is set in 1992, during the height of crime and gang activity in the sprawling metropolises of California. The player controls the main protagonist Carl Johnson, known by his nickname CJ, an African-American man approximately 25 years of age. CJ has returned to Los Santos after five years of living in Liberty City. He originally left Los Santos back in 1987 due to the death of his younger brother Brian, for which his older brother Sean Johnson, known by the nickname Sweet, blamed him. He has now come back to the city to attend the funeral of his mother, Beverly Johnson. She was killed by Sweet's enemies during a drive-by shooting.

On his way home, CJ is stopped and detained by Los Santos police officer Frank Tenpenny, the head of the special operations unit known as C.R.A.S.H.
